Mesembryanthemum crystallinum extract ameliorates TNF-α-induced inflammation and insulin resistance in 3T3-L1 adipocytes

Obesity is classified as a low-grade chronic inflammatory disease. Obesity-induced inflammation associated with the development of insulin resistance (IR). In this study, we investigated effects Mesembryanthemum crystallinum extract (MCE) on tumor necrosis factor-alpha (TNF-α)-induced and IR in 3T3-L1 adipocytes, well underlying mechanisms. Lipolysis was monitored by oil red O staining. Glucose uptake measured 2-deoxyglucose (2-DG). The production cytokines adiponectin evaluated using enzyme-linked immunosorbent assay (ELISA). Protein expression western blotting. induced differentiated adipocytes treatment TNF-α. MCE decreased lipolysis while increasing production. inhibited pro-inflammatory such interleukn (IL)-6 monocyte chemoattractant protein (MCP)-1, Nuclear factor-κB (NF-κB) translocation. Additionally, increased glucose uptake, prevented phosphorylated receptor supstrate (IRS) Transporter Type 4 (GLUT4) dose-dependent manner. These results demonstrate potential improving sensitivity obesity. Therefore, can be used novel candidate for developing functional foods.
